I looked into this briefly, and it appeared you had to go through Grand Cayman to get to Cuba from Jamaica.
Flights to Cuba will become easier and easier to access in the years to come. Air Jamaica flew to Cuba when they were around but the prices were outrageous.
I'm not sure if Cubana airlines fly to Montego Bay. Maybe check their site.
If you cannot get there via Jamaica, depending on where you live in the States you can enter through a third country such as Cayman, the Bahamas, Mexico (City) or Canada. If you want to jump through diplomatic hoops; which have become easy to jump through under the Obama administration, you can fly direct to Havana from Miami (as always) and now from New York as well.

It does not seem that Cubana has flights to Jamaica at the moment so to answer the question of the original post, no, there is no way to get a scheduled flight from Negril (Jamaica) to Cuba. That will probably change in the years to come however.
One day a couple of city slickers were out driving in the countryside and eventually got lost. They spotted a farmer working his field and thought they would stop and ask for directions. They asked the farmer for directions to such and such place. The farmer gave the question some thought, wiped his brow, and replied, "I reckon you can't get there from here."
Uncle Johnny told me that you could take a cab FROM NEGRIL *nods to rob* to Kingston, then take a cruise from there.
