Okay, a bit more information about a fecal coliform test.
It needs to be completed within 6 hours of taking the sample not 2 hours as i first stated. Once the sample is taken, it should be run right away or placed in a fridge at 4 degrees celcius for up to 6 hours. The reason it need to be run in that time frame is because if bacteria is present it will start "using up" all the oxygen. A sample that has sat for a day simply has run out of oxygen and died. In the instructions in the photo it talks about adding the "medium". Basically that is food that is added to let the bacteria grow. The bacteria need food and oxygen to grow.
