If I buy a bottle of alcohol in duty free during a US layover, do I have to “declare” it? ie use the something to declare line?

No worries. Just carry it in..Nothing to declare line

2 liters of booze per person is the rule for Jamaica. If that is all you have then you have "nothing to declare".
